Cell line
A2780
Authenticated research cell lines, mycoplasma-free and passage-controlled. Catalog number FNC0013,
part of the Ready-to-Use Cell Lines range. Send the catalog number with your
quantity and we will confirm availability, lead time and price.
Vector map
Not applicable
This catalog number is not a plasmid construct, so there is no vector map. See the handling and storage guidance below for what you receive.
How it works
Each line is expanded from a low-passage working bank, tested for mycoplasma before release and shipped with its passage number recorded. Cell line identity is the quiet variable behind most irreproducible experiments — lines drift, cross-contaminate and mislabel — so authentication and passage control are treated here as product attributes, not extras.
Lines in this catalog serve two main workflows in the viral vector space: producer and packaging substrates (HEK293T and derivatives), and permissive target models for transduction, oncolytic sensitivity and titer assays. Culture conditions differ line by line — medium, serum percentage, splitting ratio and confluence limits all matter — so always start from the data sheet supplied with the shipment.
Production & quality control
- Working banks are expanded under standardized conditions and freeze-down in a controlled-rate program before storage in liquid nitrogen vapor phase.
- Every released lot is mycoplasma-tested negative; STR profiling is available on request for identity confirmation.
- Passage number at shipment is stated on the certificate of analysis so you can track culture age in your own records.
- Shipping is on dry ice for most lines; some fastidious lines travel at ambient temperature as a recovered culture — the product sheet states which.
Storage & handling
- Cryovials: liquid nitrogen vapor phase for storage, dry ice for transport.
- Upon receipt, transfer directly to liquid nitrogen — do not hold at -80 degrees Celsius for more than a few days, as slow ice recrystallization damages viability.
- Recovered cultures: follow the medium and passaging regime on the data sheet.
Experimental protocol
-
Thaw
Warm complete medium to 37 degrees in advance. Thaw the vial in a 37 degree water bath until only an ice crystal remains (45 to 90 seconds), transfer into 9 volumes of pre-warmed medium and spin gently if your SOP requires DMSO removal, or dilute directly for hardy lines.
-
Seed
Plate at the density stated on the data sheet — typically 2 to 4 times 10 to the 4 cells per square centimeter for adherent lines. Some lines need higher density to recover; under-seeding a fragile line after thaw is the most common recovery failure.
-
Maintain
Change medium the day after thaw and every 2 to 3 days thereafter. Passage before confluence — over-confluent cultures shift phenotype, and for producer lines such as HEK293T it directly reduces transfection efficiency. Record every passage number.
-
Bank early
Freeze a working bank from the first or second passage after thaw, not from a late culture. Freeze in 90 percent serum with 10 percent DMSO at a controlled rate, then store in liquid nitrogen vapor phase. Discard the original vial lineage once the bank has passed a mycoplasma check.
Follow aseptic technique and your institution's cell culture SOPs; the steps below are general guidance and the data sheet for your specific line takes precedence.

Frequently asked questions
How many passages can I use before re-authenticating?
A practical ceiling for most lines is 15 to 20 passages from thaw, or whatever the data sheet states, whichever is lower. Beyond that, phenotype drift becomes measurable and publication reviewers may ask for an STR record.
Is the line mycoplasma-tested?
Every released lot is mycoplasma-tested negative. Re-test in your own lab after a few weeks of culture if your funding requires a documented in-house record.
Do you provide culture conditions for each line?
Yes — the data sheet shipped with each line lists medium, serum, splitting schedule and any special requirements. Ask us if anything on it is unclear.
