Viral vector platform
Retrovirus packaging
Retroviral vectors integrate into the genome of dividing cells, which makes them useful for stable transduction of proliferating lines and for applications that need predictable single-copy integration. FuBio packages retroviral vectors on request.
What Are Retroviral Vectors?
Gamma-retroviruses such as murine leukemia virus (MMLV) are enveloped viruses that carry two copies of a positive-sense single-stranded RNA genome. After entry, the genome is reverse-transcribed into DNA and integrated into the host chromosome by the viral integrase.
The integration step is what makes these vectors useful — and what sets their limit. The pre-integration complex cannot cross an intact nuclear envelope, so gamma-retroviral vectors integrate only in cells that pass through mitosis. Expression is therefore stable and heritable in proliferating cultures, while quiescent or non-dividing cells are essentially not transduced.
Retroviral Vector Packaging and Production
FuBio packages MMLV retroviral vectors with a split system: the transfer vector carries the cassette while the structural and enzymatic functions are supplied in trans, so the particles released into the medium are replication-deficient:
-
Transfer Plasmid
Carries the expression cassette flanked by the long terminal repeats, together with the psi packaging signal that directs the RNA into assembling particles. Wild-type and self-inactivating (SIN) backbones are both available.
-
Gag/Pol Packaging Plasmid
Supplies the structural proteins of the core and the enzymes for reverse transcription and integration. Providing them in trans, separate from the transfer vector, is what keeps the preparation replication-deficient.
-
Envelope Plasmid
Provides the glycoprotein that decorates the envelope and sets tropism. VSV-G is used by default because it broadens host range and makes the particle robust enough to concentrate; other envelopes can be discussed.
The plasmids are co-transfected into packaging cells and the supernatant is harvested after a short incubation. It is clarified by centrifugation and filtration and then concentrated; higher-purity material for demanding applications can be prepared on request.
Functional titer is reported in TU/mL by a qPCR-based approach, with sterility and mycoplasma testing in the standard release package and a transduction test where the vector encodes a fluorescent protein. Keep the packaged region within the MMLV cargo budget of roughly 8.3 kilobases for the titer to be guaranteed.

Production and quality control
- MMLV retroviral vectors are produced by co-transfection of packaging cells with the transfer plasmid, a Gag/Pol packaging plasmid and an envelope plasmid (VSV-G pseudotyping by default); supernatant is harvested after a short incubation period.
- Supernatant is clarified by centrifugation and filtration, then concentrated; higher-purity material for demanding applications can be prepared on request.
- Functional titer is reported in TU/mL (transducing units per milliliter) determined by a qPCR-based approach; sterility, mycoplasma and, where the vector encodes a fluorescent protein, a transduction test are part of the standard release package.
- Wild-type and self-inactivating (SIN) backbone options are available; the packaged region should stay within the MMLV cargo budget of roughly 8.3 kilobases for the titer to be guaranteed.

1
Plan the transduction
MMLV retrovirus only transduces dividing cells, so seed cells the day before infection so they are actively proliferating at 50 to 70 percent confluence on the day of transduction. Include an untransduced control and, if you will select, a kill-curve plate. Non-dividing or primary resting cells are better served by lentiviral vectors.
-
2
Thaw and handle the virus
Thaw a vial quickly in a 37 degree water bath or on ice, mix gently — never vortex — and keep aliquots on ice while working. Avoid more than one freeze-thaw cycle: each one causes a large drop in functional titer.
-
3
Transduce
Replace the culture medium with fresh complete medium containing the diluted virus. Add polybrene at 4 to 8 micrograms per milliliter only for cell types you have confirmed tolerate it. Incubate 6 to 16 hours, then replace with fresh medium if the cell type is sensitive.
-
4
Select or enrich
Start antibiotic selection 48 to 72 hours after transduction at the concentration established by your kill curve. For fluorescent reporters, enrich by FACS or keep the whole population and record the positive fraction.
-
5
Confirm the stable line
Because MMLV integrates into the genome of dividing cells, expression is stable across passages once selected. Verify expression by the reporter, flow cytometry, Western blot or qPCR, expand under the minimum effective selection pressure, and bank an early passage.
Bench guidance only: optimize multiplicity of infection, selection pressure and timing for your own cell type, and carry out all retroviral work under the biosafety conditions approved by your institution.
Why should I choose retroviral vectors for my study?
Gamma-retroviral vectors give stable, heritable expression in proliferating cultures with a small, well understood genome and a low immune profile, which makes them a straightforward choice for stable cell line work. They are also the classic tool for insertional mutagenesis and for marking dividing populations. Where your target cells are quiescent or post-mitotic, lentivirus is the right choice instead, because only lentiviral pre-integration complexes enter an intact nucleus.
What are retroviral vectors used for?
The common uses are stable transduction of dividing cell lines, hematopoietic and progenitor cell work, insertional mutagenesis screens, and long-term expression models that must survive extended passaging. They are also used to exploit the dividing-cell restriction: in a mixed culture, only the proliferating fraction is transduced.
Why do retroviral vectors only transduce dividing cells?
After entry and reverse transcription, the pre-integration complex has to reach the chromatin, and gamma-retroviral complexes cannot cross an intact nuclear envelope. They gain access during mitosis, when the envelope breaks down, so quiescent cells are essentially not transduced. In practice this means seeding cells the day before transduction so they are actively cycling, and switching to a lentiviral vector for neurons, resting lymphocytes or other non-dividing targets.
How is retroviral titer determined and how should I dose?
We report functional titer in transducing units per milliliter, determined by a qPCR-based approach, and include it in the release package alongside sterility and mycoplasma results. Because transduction efficiency depends strongly on the growth state of your cells, run a small multiplicity-of-infection series the first time rather than relying on a single number, and include an untransduced control so the assay has a baseline.
What is the biosafety profile of retroviral vectors?
The vectors are based on MMLV and are replication-deficient: the structural and enzymatic functions are supplied in trans during production, so the particles in the preparation cannot spread. They are nonetheless commonly handled at biosafety level 2 or the equivalent approved by your institution, and vectors expressing hazardous transgenes may require additional containment. Integration is permanent, so consider the insertional mutagenesis risk when designing selection and screening strategies. All products are for research use only.
How should retroviral vectors be stored and handled?
Store single-use aliquots at minus 80 degrees Celsius and avoid repeated freeze-thaw cycles, which cause a large drop in functional titer. Thaw quickly in a 37 degree water bath or on ice, mix gently rather than vortexing, and keep the aliquot on ice while you work. Add polybrene at 4 to 8 micrograms per milliliter only for cell types you have confirmed tolerate it, and inactivate waste with a validated disinfectant before disposal.
